New Application Note: High Resolution PEPperMAP® Epitope Mapping of Autoimmune Sera

PEPperPRINT is pleased to release a new application note based on a collaboration with Protagen Protein Services about the high resolution epitope mapping of systemic sclerosis (SSc; also termed scleroderma) patient sera.

Diagnosis of SSc is based on the combination of clinical and/or laboratory features e.g. SSc-related autoantibodies. The most frequently measured autoantibodies (anti-Scl70, anti-centromere like CENPA, anti-RNPIII) are only present in about 60-70% of SSc patients. By using its SeroTag^® process, Protagen identified KDM6B (Lysine (K)-Specific Demethylase 6B) as new autoantigen with a high frequency in SSc.

To characterize the autoantibody responses against KDM6B, a high resolution PEPperMAP^® Epitope Mapping with 15mer peptides and maximal peptide overlap of 14 amino acids was done by PEPperPRINT. In parallel, we mapped the epitopes of centromere protein A (CENPA) specific autoantibodies. Based on three SSc patient sera and one healthy control serum, we were able to identify two N-terminal adjacent and common epitopes within CENPA as well as a common epitope for the new putative SSc antigen KDM6B.
