New Janssen Paper based on PEPperCHIP® Peptide Microarrays


Janssen Diagnostics recently published a paper on the identification of immunodominant Onchocerca volvulus motifs in PLOS Neglected Tropical Diseases by using PEPperPRINT’s high-density peptide microarrays.

Infections with the parasite O. volvulus can cause river blindness. Since only about 80% of infected patients can be diagnosed by testing IgG4 antibodies to the parasitic antigen Ov-16, the demand for further diagnostic tools as well as the development of therapeutics and vaccines is high.

 

By using high-density O. volvulus peptide microarrays, Janssen researchers identified three immunodominant motifs in onchocerciasis patient sera. A selection of 12 peptides, 4 for each of the 3 motifs, were further analyzed by PEPperMAP® Epitope Substitution Scans to identify conserved and variable amino acid positions of each epitope in comparison to the position of the motif site. The results showed that the conserved consensus motifs were part in nearly each wild type peptide and that the identified peptides might be candidates for immunodiagnostic applications.
